Raji TK- Cells

The Raji TK- cell line is a derivative of the Raji Burkitt?s lymphoma cell line, characterized by its lack of thymidine kinase (TK) activity. This deficiency results from the loss of the thymidine kinase gene function, rendering the cell line resistant to certain nucleoside analogs and drugs that rely on TK activity for activation. This property makes Raji TK- an invaluable tool in genetic selection experiments, particularly in studies utilizing the hypoxanthine-guanine phosphoribosyltransferase (HGPRT) salvage pathway for selective growth conditions. Like its parental counterpart, Raji TK- cells retain features of Epstein-Barr virus (EBV) infection and express B-cell surface markers such as CD19 and CD20. The cell line has been extensively used in virology, oncology, and immunology research to study EBV-mediated transformation, lymphoma progression, and immune evasion strategies. Its TK- phenotype allows for the creation of hybridomas and targeted gene-editing experiments through the selection of cells lacking TK-dependent pathways.
